http://neurophys.gu.se/english/departments/physiology/metabolic-physiology The primary goal of this project is to clarify mechanisms that underlie the deterioration of pancreatic islet function in type-2 diabetes. We are interested in electrical intracellular and intra-islet communication processes and intracellular stimulus-secretion coupling in the hormone secreting cells of the islet in diet-induced and genetically driven obesity and diabetes. A mouse model allowing for detection of intra-islet communication processes, a beta-cell specific Channelrhodopsin-2 expressing mouse line, has been generated and a second one is in preparation. This is expected to help identify the processes contributing to hypoinsulinaemia and hyperglucanonaemia, both causing hyperglycaemia, the major feature of type-2 diabetes. We are using molecular biology methods (cloning, (q)PCR, cell culture, transfections, ELISA), electrophysiology (?Patch clamp?), state-of-the-art imaging (fluorescent confocal microscopy, TIRF) and innovative Optogenetics methods to investigate pathophysiological mechanisms in mouse, rat, and clonal (eventually human) islet cells. Specific tasks will (but are nor limited to) include: - Sub-cloning and in vitro testing of vectors suitable for genetically-encoded cell-specific detection of the intra-cellular calcium concentration (GECls) and/or membrane potential (GEVIs), respectively) and preparation according to needs of collaboration partner. - Characterisation of the mouse model(s). - Mouse breeding, in vivo pharmacological treatment of rodents, glucose-tolerance tests and tissue isolation. - In vitro functional analysis of pancreatic islets and if relevant other metabolically important tissue (brain, muscle, liver, adipose). - Analysis of results and writing of reports and manuscripts. - Method demonstrations for undergraduate students. - Shared lab-duties according to schedules. The successful candidate will be able to work independently, self-motivated, analytical, and have an eye for detail. A good command of English (both written and spoken) is required. We are an international and multilingual laboratory and additional language skills are also desirable. Previous experience with Molecular Biology techniques, electrophysiology and/or imaging are merits. The 4-year PhD programme at Sahlgrenska Academy includes compulsory and elective courses (see

Active participation in in-house seminars and visiting national and international conferences, laboratories and workshops is also part of the training. The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral studentship.

Eligibility: To be admitted to third-cycle studies you must fulfill both the general and the specific entry requirements. The general entry requirements is fulfilled if you have: 1. completed a degree at a second-cycle level, or 2. completed at least 240 credits worth of studies of which at least 60 credits must be at a second-cycle level, or 3. acquired equivalent knowledge in some other way in Sweden or abroad. The specific entry requirements is fulfilled if you have completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies.
