Description of Opportunity:
We have an exciting opportunity for a biochemistry/drug metabolism scientist to join the Cardiovascular/Gastrointestinal or Respiratory and Infection (CVGI or R&I) Innovative Medicines (iMED) DMPK departments. There you will study the in vitro formation, reactivity and biological effects of drug carboxylic acid - CoA conjugates in model systems to assess toxicity and/or covalent binding as biochemical endpoints.
The focus of your work will be on analysis of formation and chemical reactivity of acyl CoA conjugates, and evaluation of their ability to cause toxicity in cell-based systems in vitro, in comparison with acyl glucuronides. You will develop robust and reproducible in vitro systems for the generation of CoA esters for a wide range of model substrates, and establish analytical methodology and model systems where reactivity can be demonstrated. Preclinical models in vivo using e.g. the hepatic reductase null (HRN) mouse will also be evaluated. The aim of your work will be to improve mechanistic understanding of carboxylic acid drug toxicity to aid in the discovery of drug candidates with reduced propensity to cause idiosyncratic adverse drug reactions in man, and in their risk assessment.
Your scientific research will also give you the opportunity to work collaboratively with other experts within DMPK, as well as fellow scientists across other disciplines, e.g., Medicinal Chemistry groups and Global Safety Assessment, while at the same time being able to work independently. You will be expected to deliver the results of your novel research in-house via global working groups and externally via meetings and through publications.
Minimum experience:
A PhD in one or more of the following areas:
Chemistry of drug metabolism
Enzymology of drug metabolism
Analytical toxicology/biochemistry; mass spectrometry
Endogenous lipid biochemistry
