Dina arbetsuppgifterType of employment: Fixed-term employment, 4 years Extent: 100 % Location: Sahlgrenska Academy, Gothenburg First day of employment: As agreed Reference number: UR 2016/19 Job assignments: Bacteria may use biofilm formation to evade elimination by host phagocytes and antibiotic treatment, a phenomenon contributing to disease development and progression in situations where a foreign body, such as an implant, has been introduced in the host. Our hypothesis is that S. aureus bacteria use activation of the host's own fibrinolytic system to digest biofilm components and be released into the tissues and circulation. Our main aim is to clarify and target molecular pathways involved in fibrinolysis and biofilm formation to interfere with S. aureus virulence and ultimately develop drugs to achieve this interference. This PhD project contains several sub-projects that will to some extent be run in parallel. S. aureus strains from patients and healthy controls will be compared with respect to expression of staphylokinase (a staphylococcal fibrinolysis activator). On the host side, activation of fibrinolysis will be analyzed in the surrounding micro-environment of the tissues (patients with indwelling device infections). Selected animal models will be used to delineate the role of fibrinolysis during cathether-associated infection. We will analyze the mechanisms whereby antibiotics as well as host defending phagocytes are shut out from the infection by the biofilm. Finally, we want to target fibrinolysis to prevent biofilm infections by pre-coating foreign medical devices with fibrinolysis activators. The planned project enables a translational approach, bridging population based infectious epidemiology and molecular studies of the microbe and host, which will lead to better understanding o f the pathogenesis of biofilm-associated disease and enable us to develop new concepts for therapeutic intervention. The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral studentship.
Utbildning och erfarenhetEligibility: To be admitted to third-cycle studies you must fulfill both the general and the specific entry requirements. The general entry requirements is fulfilled if you have: 1. completed a degree at a second-cycle level, or 2. completed at least 240 credits worth of studies of which at least 60 credits must be at a second-cycle level, or 3. acquired equivalent knowledge in some other way in Sweden or abroad. The specific entry requirements is fulfilled if you have completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies.
