Absorption from the gastrointestinal tract is the first critical step for oral drug treatment. The general principles of intestinal permeability are quite well understood, and can be predicted with in-silico models or from in-vitro cell culture systems, however metabolising enzymes and efflux transporters are known to be a permeability limiting factor in the absorption of new drugs.
As a complement to traditional in vitro systems, the ADME team within Early CVRM DMPK can access fresh rat and human intestinal tissue to study drug absorption and metabolism using a sophisticated ex-vivo Ussing Chamber system. This ex-vivo system represents the complex in vivo situation owing to the maintained morphological and physiological features of the intestine such as functional transporter and enzyme systems.
Current in vitro transporter assays require the use of transporter-specific compounds as substrates or inhibitors to identify potential drug-transporter interactions. However, substrates are often recognized by multiple transporters at different affinities, and the specificity of inhibitors is often unknown or poor, leading to ambiguous interpretations of drug-transporter interactions.

Responsibilities
We offer a student placement to perform their MSc-thesis in this exciting and important field in our ADME team (Early CVRM DMPK) with a focus on validating the ex-vivo Ussing model for transporter studies of the two main efflux transporters, BCRP and P-gp.
Qualifications
The placement offers a possibility to gain an in-depth understanding of advanced in vitro studies of drug intestinal absorption and metabolism in a project setting. Being integrated in the ADME team, the student will learn more about ADME principles and the impact of ADME in other organ systems and its relevance to the drug development process.
This placement is connecting multiple scientific skills, and the student will have opportunity to integrate in-vitro work with fresh intestine, bioanalysis of own samples with LCMSMS, and building knowledge on drug transporter kinetics, and data evaluation. The results will be compared to published clinical absorption data.
