Project title:
Defining the biochemical and medical importance of GGTase-I
Job assignments
Our goal is to understand the biological significance of the posttranslational processing of CAAX proteins (e.g., RHO proteins and prelamin A) and to define the importance of the CAAX processing enzymes in the pathogenesis and treatment of inflammatory disorders and progeria. The C-terminal CAAX motif undergoes three processing steps by four different enzymes (i.e., FTase, GGTase-I, RCE1, and ICMT). It is widely believed that these processing steps are essential for the membrane targeting and activity of CAAX proteins.
Our recent studies challenge this dogma. Knockout of GGTase-I, the enzyme responsible for geranylgeranylation of RHO proteins, hyperactivated RHO proteins, did not affect membrane targeting, and induced rheumatoid arthritis in mice. Evidently, geranylgeranylation inhibits, rather than stimulates, RHO protein activity. We now need to re-evaluate the importance of geranylgeranylation, define pathways that drive arthritis developme nt, and determine if this mechanism contributes to human arthritis.
In this PhD-project, the successful candidate will use mouse genetics, in vivo conditional and conventional gene targeting, a wide range of cellular and molecular biology techniques to address several specific aims. The candidate will also work alongside other investigators on one or two side projects.
PhD positions are financed mainly through education grants during the first two years and employment as doctoral students during the last two years and pertains to a docotral degree
Eligibility
The general entry requirements are fulfilled if you have either 1, a second cycle degree (usually described as a masters degree) or 2, completed courses at an academic level corresponding to at least 240 education credits. At least 60 of these credits must be at second cycle level. 60 credits is equal to one year of full-time studies. You also need to fulfill the specific entry requirements which is an English language requirement; Engelska B.
Assessment
After the application deadline has passed, an initial assessment of the candidates eligibility is made at the faculty office and sent to the project supervisor for selection. The process can then take up to 6 months before a candidate is selected.
