Regulation of metabolic and inflammatory profile in peripheral tissues
Application deadline: 2012-02-09
Description of the research project
Type 2 diabetes (T2DM) is characterized by hyperinsulinaemia, hyperglycaemia and dyslipidaemia. With its steep increase in incidence, T2DM is becoming one of the major global threats to human health. In industrialized countries, the total burden of diabetes and related complications is approximately 10% of the total health care costs. Since the efficacy and patient compliance of existing treatments are in many cases unsatisfactory, there is a pronounced need for novel targets, which could complement current treatment.
Several lines of evidence provided by us as well as other research groups suggest that by activation of AMP-activated protein kinase (AMPK) one can increase glucose uptake and fat oxidation in the skeletal muscle, inhibit glucose output and triglyceride synthesis in the liver and reduce body fat content, thus reverting the main metabolic disturbances in T2DM. Our most recent studies also demonstrate a key function of AMPK in suppression of inflammatory reactions caused by the cytokine action in liver, suggesting that AMPK may act as an intracellular link between chronic inflammation and metabolic regulation in peripheral tissues. The aim of the project is to elucidate the signalling pathways for AMPK to control metabolic balance in peripheral tissues. The study will contribute to the understanding of the complex and integrated molecular signalling mechanisms regulating the whole body metabolic profile and of the changes leading to obesity, T2DM and cardiovascular disease. Importantly, the project will potentially reveal novel approaches in therapeutic intervention of the metabolic syndrome and related complications.
