Thymic exosomes in T cell development

Description of the research project
This PhD project aims at investigating the role of exosomes as communicators between different cell populations in the thymus. Exosomes are membrane bound nano-vesicles that are produced by various cell types and have been shown to allow communication between immune cells both by carrying antigen and functional RNA. During T cell development in the thymus the thymic epithelial cells and the dendritic cells communicate intensely with the developing T cells in order to shape the T cell repertoire and function. We believe that this communication involves the production of informative exosomes. The project breaks down into the following specific aims, which will correspond to the publications building the thesis:
• To study the formation and release of exosomes from human thymic explants and analyze their morphology and protein composition.
• To analyze the RNA and microRNA content of thymic exosomes.
• To monitor the production and function of exosomes derived from different thymic cell populations.
• To study the impact of thymic exosomes on T cell development and their ability to induce regulatory T-cells.
• To study thymic exosomes from patients with genetic alterations associated with a high risk for the development of immune deficiencies and/or autoimmune disorders.
The project involves both structural and functional studies using confocal and electron microscopy, proteomics, genomics and advanced immunological techniques. The project combines in vitro studies on human thymus and clinical studies on immuno deficient patients combined with experimental in vivo studies that involves genetically modified mouse strains.
