Understanding the regulatory requirements for gut IgA B cell responses and their potential impact on mucosal vaccine development

Description of the research project
Mucosal immunology is attracting increasing attention. The formation of IgA antibodies is a particular feature of mucosal immune responses and is unique to B cells residing in mucosal tissues. However, little is known about the processes that determine IgA B cell development and differentiation. Disappointingly few oral or mucosal vaccines have been developed and none are available using subunits or recombinant antigens from pathogens. This proposal aims at changing this situation by developing a better understanding of IgA B cell responses and long- term memory development. We have developed a mouse model based on a NP-haptenated cholera toxin conjugate that allows us for the first time to study phenotypically distinct and easily identifiable memory B cell populations, and analyze their expansion and distribution in the immune individual following a challenge with antigen. This allows us to sequence the data from isolated IgA B cells from various mucosal tissues and GALT to determine clonal relationships and the level of acquired mutations, and also to follow individual B cells involved in the immune response. The project represents a shift in paradigm and will have great impact on our thinking about mucosal immunomodulation and vaccine design. In addition, it will unravel the regulatory mechanisms governing mucosal IgA immunity and memory B cell development. The PhD student will be part of a research team within MIVAC, a center-of-excellence, consisting of 5 members, with Dr Mats Bemark and Prof Nils Lycke as co-supervisors, who work with various aspects of mucosal IgA B cell responses and immunomodulation. The group is internationally well recognized and has made some pioneering contributions to the field of mucosal IgA B cell immunology and vaccine design published in three recent issues of The Journal of Immunology.
Desired background
A background within molecular biology, immunology or microbiology is desired. Previous laboratory experience within these areas is advantageous.
