Dina arbetsuppgifterType of employment: Fixed-term employment, 4 years Extent: 100 % Location: Sahlgrenska Academy, Gothenburg First day of employment: As agreed Reference number: UR 2015/903 Project title: Immune regulation of gut IgA immunity and oral tolerance using a toxin-adjuvanted nanoparticle vaccine approach. Job assignments: Although it is commonly agreed that mucosal vaccines would dramatically improve global health only a handful of mucosal vaccines exist today. The reason for this is lack of effective immunoenhancers (adjuvants) and delivery systems for clinical use. Here we combine interdisciplinary expertise in generating a generic mucosal vaccine platform by combing novel lipid nanoparticle (LNP) technology with a unique adjuvant-targeting strategy based on the CTA1-DD adjuvant, a non-toxic ADP-ribosylating derivative of cholera toxin. Already successful experiments have shown this combination to be an effective mucosal vaccine platform that stimulated protective immunity. The PhD project will now study targeting and modulation of function in mucosal dendritic cells (DC) after oral delivery of the combined LNP vectors. A detailed understanding of the regulatory environment in the mesenteric lymph nodes (MLN) and Peyer's patches (PP), including subsets of CD4 T cells and DCs, will be undertaken with in-depth investigations of signalling pathways in DCs responsible for the mechanism of action of the CTA1-DD adjuvanted LNP vaccine. The latter involves studies of the role of Gsa and other potential G-proteins in DCs, which are the targets of the CTA1-DD adjuvant in DCs. Hopefully, this work will lead up to that a mechanistic distinction can be made at the molecular level between an immunoenhancing and a tolerogenic response. Moreover, how these effects on DCs and CD4 T cell subsets can influence IgA B cell responses in PP will also be evaluated using two-photon microscopy and sophisticated cell-specific reporter mice. The project is both timely and unprecedented in its design and strength, as mucosal vaccines against infections or treatments against autoimmune diseases are much warranted. An interdisciplinary team of internationally renowned researchers with a documented track record of successful collaborations has bee n brought together to undertake the work at MIVAC. The PhD project will be part of a larger project that I am the principle investigator of and which has attracted financial support from EU, VR , Cancerfonden, KAW and SSF. We would like the applicants to motivate why they interested in the project and why they are suitable applicants. The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral studentship.
Utbildning och erfarenhetTo be admitted to third-cycle studies you must fulfill both the general and the specific entry requirements. The general entry requirements is fulfilled if you have: 1. completed a degree at a second-cycle level, or 2. completed at least 240 credits worth of studies of which at least 60 credits must be at a second-cycle level, or 3. acquired equivalent knowledge in some other way in Sweden or abroad. The specific entry requirements is fulfilled if you have completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies.
