The research area and duties

The project is part of our efforts to clarify the role of cooperative phenomena and hierarchical ordering in accounting for high power-output and resistance to stretch of striated muscle in health and disease (e.g. hereditary cardiomyopathies). The project will be carried out in collaboration between A. Månsson and S. Tågerud at Linnaeus University, H. Linke (Lund University, Sweden), D. Rassier (McGill University, Canada) and J.A. Spudich (Stanford University, USA). The focus will be on the development and use of a new experimental set-up for in vitro measurement of forces developed by small ensembles of isolated myosin motors as different length changes are imposed on actin filaments under different conditions (mutated myosin, presence of drugs, etc.). Modelling of the actin-myosin interaction for in-depth interpretation of the experimental results may also be part of the post-doc project. The work will be performed mainly in Kalmar, but extensive interaction with labs of collaborators is expected. The postdoc, whose main focus will be on instrumentation, will work in close collaboration with a PhD student with a focus on biochemistry and biophysics of isolated muscle proteins. Key experimental techniques, in addition to the newly developed force-measuring device, will be fluorescence microscopy and in vitro motility assays.

Qualifications

Applicantsshould have a PhD degree in a subject area of relevance for the project (biophysics, computer science, physics, engineering, etc.). Experience of experimental research work using fluorescence microscopy and biophysical techniques for recording of force and length changes from cells or proteins would be an advantage. The same applies to previous experience of mathematical modelling and development of new experimental techniques. Previous studies of molecular motors and muscle are judged favourably.
