The Neurochemistry laboratory at the Institute of Neuroscience and Physiology, at the Department of Psychiatry and Neurochemistry, conducts neurochemical research targeted to understand the pathophysiology of Alzheimer's disease and other brain disorders. An important goal is to identify protein changes linked to the pathophysiology, known as biomarkers, which can be valuable to explore disease mechanisms, for diagnosis and to monitor treatment effects.
We are looking for a motivated and ambitious postdoc fellow with background in biochemistry and molecular biology to investigate post-translational modification profiles of tau (e.g., aggregation, phosphorylation and truncation) in multiple tauopathies using an exploratory approach. The aim is to identify molecular pathological signatures that are common to all tauopathies and others specific to different diseases.

Subject area description
Alzheimer's disease (AD) is the most studied dementia. The pathological hallmarks of AD include brain accumulation of: (i) amyloid plaques consisting of aggregated amyloid beta (A) peptides and (ii) neurofibrillary tangles made of phosphorylated tau (p-tau) aggregates. These pathological changes are reliably measured in cerebrospinal fluid (CSF) using immuno-assays including those developed in our lab. Such biomarker measures are widely used to support clinical diagnosis and candidate drug testing. Recently, our lab discovered p-tau and A biomarkers that work well in blood.
However, current CSF and blood biomarkers are increased specifically in AD; yet, levels in other tauopathies are same as in controls. Moreover, CSF and blood total tau, a marker of neuronal injury, and novel tau fragments modified in AD are not altered in other tauopathies despite the presence of severe tau pathology and neuronal injury. Other biomarkers, including those that monitor neuronal degeneration (e.g., neurofilament light, neurogranin), similarly do not differentiate AD from non-AD tauopathies.
Consequently, identifying and distinguishing between non-ADs with shared clinical features remain a challenge that this project aims to address.
Job assignments
The postdoc fellow will investigate post-translational modification profiles (e.g., aggregation, phosphorylation and truncation) of tau protein in AD and related tauopathies. The postdoc will work to identify molecular pathological signatures that are common to all tauopathies and others specific to different diseases. The tools to be used include mass spectrometry of proteins (proteomics), immunoprecipitation (IP), and immunoassay technologies (e.g., ELISA, Simoa, and similar). The postdoc will perform these studies using well-characterized brain tissue, CSF and blood samples from clinical sources. This knowledge will then be applied to develop and validate novel biomarker tools for diagnostic and prognostic use. The project will combine discovery/exploratory and targeted approaches to maximize the likelihood of success.
Eligibility
To qualify as a postdoctoral researcher the applicant should hold a PhD in a relevant subject. Since postdoctoral constitutes a qualifying appointment for junior researchers, we aim to target those who have a doctoral degree not older than 3 years from the application deadline.
Assessment
Candidates will be assessed based on documented scientific experience with focus on method development and knowledge with regard to biochemical methods such as immunoassays and mass spectrometry of biological samples, as demonstrated by publications in international, peer-reviewed journals. Previous experience in assay development using Simoa or related platforms, mass spectrometry, or working with brain tissue, CSF or blood samples or similar. Demonstrable experience in neurochemical biomarker development for brain pathologies is a particular merit.
Ability to independently process data and write scientific publications is essential, as is the ability to be flexible and able to work independently. During assessment, account will be taken with respect to personal ability and qualifications. Very good proficiency in English is mandatory.
Employment
The employment is limited (temporary) for 24 months, 100% of full-time, and is placed at the Institute of Neuroscience and Physiology. Starting date; as soon as possible or as agreed.
