This project is a part of NextBioForm consortium that involves several universities and companies. The consortium aims at building a well-renowned, globally competitive sustainable national centre for development of biologics. In this project, we will investigate the structural and thermodynamic changes in proteins upon dehydration in the presence of pharmaceutical excipients. Furthermore, we plan to study structural changes in semi-dry protein formulations that occur upon heating to temperatures that typically lead to protein denaturation. The main techniques involved in the project will be thermal analysis and water sorption techniques such as DSC, sorption calorimetry and humidity-scanning QCM-D. Together with synchrotron SAXS (CoSAXS beamline at MAX IV) they will be used to develop understanding of mechanisms of destabilisation of proteins in the dry state and the effects of excipients.
The CoSAXS beamline is a state-of-the-art multipurpose Small Angle X-ray Scattering (SAXS) instrument at MAX IV which is currently under construction. It will offer high brilliance, monochromatic and tuneable X-rays, with outstanding performance in low beam divergence, high X-ray flux, and variable beam size in both directions in the full range of the sample-detector distances provided by the beamline (up to 18 m). Several techniques will be available through modular operation: time resolved SAXS, SAXS/WAXS, protein solution SAXS, microfocusing SAXS and XPCS.
Apart from carrying out research within these areas the applicant is expected to take responsibility for the maintenance of the instruments involved and to be active by supervising postgraduate students and fellow scientists in the techniques, as well as participate in teaching at post graduate and graduate levels. Teaching at undergraduate level may also to some extent be required. In addition, the applicant should (when applicable) be responsible for contacts with involved industry partners and is also expected to take part in applications for external funding to support research activities.
Candidates must be physical chemists or biophysicists by training and have a doctoral degree together with a good track record in surface and colloid chemistry or pharmacy. Special considerations will be given to scientific expertise in and previous hands-on experience in the following methods: DSC, synchrotron-SAXS (including modelling), SWAXD, microcalorimetry, ITC, TGA, DVS, QCM-D. Documented excellent level of English (understanding, speaking and writing) is essential. The candidate should also have the ability to work in a team and collaborate with people in complementary disciplines. Previous experience from working in research projects that involved both industry and academia is highly valued.
The position is placed at the Faculty of Health and Society, Department of Biomedical Science. The education and research activities comprised by the faculty aim at contributing to the knowledge base in order to preserve, support and promote people's physical, psychological, and social health. Research is taking place in areas ranging from science, technology and medicine to social science. At the interfaces between them, exciting new areas for research are being generated. The research at the faculty is also linked to the research programme Biofilms - Research Center for Biointerfaces which combines research with medical, dental, food and environmental applications, at three of our faculties, in close collaboration with regional industry
