Titel: PhD student position in medical science - Studies of genetic alterations related to clinical outcome for patients with triple negative breast cancer

Type of employment: Fixed-term employment, 4 years if full-time studies Extent: 100 % Location: Sahlgrenska Academy, Gothenburg First day of employment: As agreed Reference number: UR 2015/517 Adjuvant treatment regimens for breast cancer are primarily based on patient- and tumor-related factors, e.g. patient menopausal status, tumor stage and histological grade, and the status of molecular tumor markers (HER2/neu and the estrogen receptor). Despite improvements in survival rates, about 20% of breast cancer patients experience recurrence within five years of initial therapy. There is therefore a need to improve patient risk assessment and to personalize therapy according to a combination of patient-specific clinicopathological features and tumor characteristics. The project goals are to analyze primary invasive TN tumors using high-throughput RNA sequencing to identify mutations and gene expression variations which are related to clinical outcome for triple negative breast cancer patients. Potential candidate genes identified using this method will be used in functional studies and analyzed at the protein level using immunohistochemistry in larger TN breast cancer cohorts. Specifically, the objectives are to (1) identify mutations and define differentially expressed genes resulting in different clinical outcomes for TN breast cancer patients, (2) activate and inhibit the function of candidate genes by transfecting different breast cancer cell lines, (3) study the oncogenic potential of the candidate genes through injection of the transfected cells in nude mice, (4) define a panel of biomarkers for immunohistochemistry that will be validated by tissue microarrays in independent cohorts of TN breast cancer patients. The goal is to determine if these biomarkers could subsequently provide diagnostic probes for TN tumors. Experimental work will include cell culture, animal handling (nude mice), drug therapy of cells and animals, and molecular biology techniques, e.g. real-time PCR, RNA interference, RNA sequencing, immunohistochemistry, etc. Associate Professor Khalil Helou's research group consists of senior researchers (geneticist Khalil Helou, oncologist Per Karlsson, pathologist Anikó Kovács), technical assistants (May Semaan, Shahin De Lara), PhD students (Luaay Aziz, Elisabeth Werner Rönnerman), and post docs (Toshima Parris). In addition, Dr. Helou's group also has a well-established collaboration with other cancer researchers including oncologists (Barbro Linderholm, Zakaria Einbeigi), biostatistician (Szilard Nemes, PhD), pathologist (Göran Landberg), molecular biologist (Jeanette Nilsson CMB). The project will be conducted at the Sahlgrenska Cancer Center. We would like the applicants to motivate why they are interested in the project and why they are suitable applicants. The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral grant during the first year and doctoral studentship during the last three years, alternatively doctoral studentship.

Eligibility: The general entry requirement is fulfilled if you have either a second cycle degree or have completed courses at an academic level corresponding to at least 240 credits. At least 60 of these credits (corresponding to 1 year of full-time studies) must be at second cycle level. You also need to fulfill the specific entry requirement which is an English proficiency requirement equal to the Swedish course English B/6.
