Studies of host glycoconjugate receptors for Helicobacter pylori and Vibrio cholerae

Description of the research project
The adhesion of pathogens and their toxin to target cells is an essential early step in infection and the majority of known microbial attachment sites on host cells and tissues are glycoconjugates. The primary goal of the present project is to elucidate the molecular basis of the bacterial-host crosstalk mediated by protein-carbohydrate interactions which is essential for directing Helicobacter pylori and Vibrio cholerae to their target tissues and for the induction of disease.
The work plan contains two parts:
1) Identification of glycoconjugate receptors: Binding assays, isolation and structural characterization of binding-active glycoconjugates by mass spectrometry and NMR.
2) Neutrophil activation studies: Chemiluminescence studies with mutant H. pylori strains lacking potential virulence factor, and in the presence of receptor analogues, or inhibitors of intracellular signaling pathways.
The ultimate goal is to create a basis for development of receptor saccharide analogues for inhibition of the initial attachment of the bacteria to their target cells, as a novel form of anti-infection therapy.
Desired background
It is desirable that the candidate has background in biochemistry or molecular biology. Previous experience of glycosphingolipid preparations and structural characterization with mass spectrometry and NMR are advantageous.
