The doctoral student positions are financed mainly through education grants during the first two years and employment as doctoral students during the two last years, alternatively employment as doctoral students during four years, and pertains to a doctoral degree.

Experimental and clinical studies on the importance of inflammation for behaviour and psychiatric morbidity

Background
Growing evidence suggests that inflammation is of importance for the patho physiology of psychiatric disorders. The major aim of this project is to increase the know ledge of the aetiology of psychiatric disorders by studying the influence of inflammation on dif ferent aspects of brain functions. Our hypotheses are

i) that inflammatory mediators are highly significant for mental functions by modulating e.g. neurotransmission and/or the development/plasticity of the brain,
ii) that genetic variations in genes of importance for the immune system hence may contri bute to inter-individual differences in behaviour and susceptibility to mental illness, and
iii) that aberrations in the immune system may contribute to the marked co-morbidity be-tween certain psychiatric illnesses and cardiovascular diseases.

We have previously shown e.g that the well-known marker of inflammation C-reactive protein (CRP) is found in raised levels in individuals with anxiety-like personality traits, that genetic variability in the gene encoding CRP is associated with impulsiveness in a healthy population as well as in a cohort of suicide attempters. Moreover, the genotype associated with impulsiveness was more common among suicide attempters than in healthy controls. This is of interest since impulsiveness is an established risk factor for suicide. Moreover, we have reported the influence of other genes of the immune system on e.g schizophrenia and personality traits not observed earlier. The underlying mechanisms for these associa tions as well as other issues with respect to inflammation, behaviour and mental illness will be studied in this project.

Description of the research project
In order to explore our hypotheses we aim to combine ani mal and human studies. Firstly, an animal model of depression will be used to study the expres sion of inflammation markers in brain in combination with treatment and behavioural experi ments and studies regarding neural plasticity and neurogenesis. Secondly, studies of variability in candidate genes in a cohort of bipolar patients will be conducted and analysed in comparison to controls as well as with respect to e.g. cytokines in cerebrospinal fluid and grey matter volume. Also, neuropsychiatric symptoms in children recruited by the Twin Registry will be studied in relation to these genes. We will also pursue a clinical study that will shed light upon the effects of anti depressant treatment on inflammation and related parameters.
