hela Sverige Long title: Exploring systemic modulation of lipid nanoparticle (LNP) uptake and efficacy: can we use physiological knowledge to make organisms more receptive to nanomedicines and nanomedicines more effective?

Pharmaceutical Sciences is a global organisation that is responsible for engineering delivery systems for therapeutics. This ranges from familiar technologies like pills and solutions, to more advanced preparations for inhalation and nanomedical applications.
New pharmaceutical modalities are being developed that expand the potential target space well beyond what is possible with small molecules. Many of these new modalities are, unfortunately, relatively large and cell-impermeant, necessitating the use of delivery systems.
Limited work has been done to understand how the systemic state and local cellular environment can affect responses to nanoparticles and, how the particles are affected by the recipient organism. An organism's metabolic state, nutritional status and hormonal balance can affect how the particles are
modified once present in the body, how they are taken up by organs and cells and, how they are processed intracellularly.
You will build on the results of internal experiments that have revealed some interesting possibilities. Your unique experience and expertise will be used to meet the following project objectives:
• Reproduce, expand and improve on our preliminary findings that show we can improve the efficacy of nanomedicines by manipulating physiological states.
• Confirm translation from in-vitro to in-vivo, and from animal to human (pre-clinical).
• Characterize the mechanisms that underlie efficacy improvements.
• Apply this mechanistic knowledge and demonstrate an improvement in the efficacy of nanomedicines, both in-vitro and in-vivo.
• Communicate our findings through publications and best practice recommendations for in-vitro and in-vivo studies of nano medicines with respect to experimental conditions and design.
How the physiological state of cells and organisms can affect the efficacy of large molecule formulations, and mRNA formulations in particular, is not well studied. This work will be of broad interest to the field of nanomedicines and is of central importance to AstraZeneca. Nanomedicines themselves are currently of great interest because the industry is embracing pharmaceuticals that are larger than traditional small molecule drugs and this requires delivery systems. Following this post-doc the applicant will have an attractive skill set and be well-positioned for continuing within the industry or academia.
You will be embedded in a multidisciplinary team, that includes other post-docs, where you will have the opportunity to learn from considerable internal expertise with currently relevant technologies including precise genome editing, proteomics and high content biology. You will also be exposed to experts working on developing cutting-edge drug delivery technologies. Our expertise and collaborative atmosphere will support you in producing high impact work that you will publish and present to the external international scientific community.
This is a 3 year programme - 2 years will be a Fixed Term Contract, with a 1 year extension which will be merit based. The role will be based in Gothenburg with a competitive salary on offer.
Essential skills and capabilities
• Ph.D. degree with specialisation in physiology, biotechnology, cell biology or equivalent scientific discipline.
• You should have a passionate relationship to science, a collaborative attitude and, the desire to help create the best working environment possible.
• Preferred candidates will have experience with in-vitro cell culture, in-vivo experiments, nanoparticle formulation and, human-derived samples.
• Laboratory automation, high-content biology (robotic microscopy and image analysis), biomarker analysis, proteomics and lipidomics (mass spectrometry) and, gene editing technologies, will be used. Preferred candidates will have a high technical aptitude and be able to demonstrate experience in most or all of these areas.
• Good knowledge of endocytosis and intracellular trafficking pathways.
• Proven ability to drive innovative research though internalisation of new techniques and technology, as well as forming internal and external collaborations.
• Track record of scientific excellence, with a strong publication record and engagement with the scientific community through both conferences and collaborations.
Desirable
• Experience with?endocytosis and intracellular trafficking, including quantitative evaluation and signal transduction.
• metabolic syndrome, the pathobiology of obesity or diabetes and/or relevant animal models of physiological states related to the dysregulation of nutrient homeostasis.
• model systems for studying lipid physiology and homeostasis and/or carbohydrate physiology and homeostasis, such as glycoprotein biology.
• robust assay development, including multivariate data analysis, processing and visualisation and, statistical tools for factorial experimental design.
• drug delivery technologies for the intracellular delivery of macromolecules.
• Excellent communication skills in english (written and spoken).
• Supervisory skills and experience (for students and more junior personnel).
