Sahlgrenska akademin utlyser doktorandplatser med placering vid institutionen för biomedicin, institutionen för kliniska vetenskaper, institutionen för medicin och institutionen för neurovetenskap och fysiologi.
Doktorandplatserna avser två års utbildningsbidrag och två års doktorandanställning, alternativt fyra års doktorandanställning, och beräknas leda fram till doktorsexamen.

Lipotoxicity in myocardial infarction and heart failure. Importance of endogenous lipoproteins for cardiac function, structure and arrhythmias

Description of the research project
The aim of this project is to test the hypothesis that endogenous production of apoB lipoproteins in the heart is an important cardioprotective system which prevents/attenuates negative consequences of intracellular lipid accumulation during development of myocardial infarction (MI) and heart failure (HF)
Using well-characterized small-animal models complemented with in vitro experiments in cell cultures we will address the association between development of cellular lipotoxicity and disturbances in myocardial energy metabolism during of acute MI and HF. To test our hypothesis we will use 31P and 1H magnetic resonance spectroscopy techniques for in vivo animal studies for non-invasive studies of myocardial lipid and energy metabolism. We will address the hypothesis that a new form of cardiomyopathy- takotsubo cardiomyopathy ("broken heart syndrome") - is a special case of acute lipotoxic heart disease.
Our preliminary results from animal studies including experiments from transgenic mice strongly support the hypothesis that myocardial apoB may be a major cardioprotective system that may influence survival in the setting of MI and HF.
The knowledge about the mechanism involved in development of disturbed myocardial energy metabolism and of lipotoxicity may contribute to define new pharmacological targets for treatment of MI and HF.
