Cholera remains an important global health problem with an annual incidence of 3 million cases and 100,000 deaths. Effective oral cholera vaccines have been developed, especially the Sweden-developed Dukoral vaccine and its recent Indian descendent Shanchol, but these vaccines are produced in insufficient amounts and at too high price to cover global vaccine needs, especially in poor developing countries.
Development of fermentation methods for preparation of vaccine strain and of inactivation methods leading to killing of the bacteria without loss of vaccine properties. Development of optimal fermentation conditions for maximal production of rCTB. Collaborate in the development of tablet/capsule vaccine formulations. Establishment and validation of of standard operating procedures (SOP) for qualitative and quantitative characterization of of cholera vaccine strain, rCTB and vaccine formulations. Preclinical vaccine studies of the new vaccines compared to licensed vaccines (Dukoral and Shanchol) with determinations of local immune responses in the intestine and serum antibody responses after oral cholera vaccination. Basic research to further define the role of the wbeT gene in Vibrio cholerae.
An achieved doctoral degree is compulsory. The doctoral thesis shall be in a relevant area according to the specific position stated here. Documented experience of molecular biological, biotechnical, immunological and animal experimental research on cholera bacteria, cholera toxin proteins and oral cholera vaccine and demonstrated potential for successful research and development on the set job assignments.
