Beskrivning av arbetsuppgifter:
Enterotoxigenic Escherichia coli (ETEC) bacteria are the most common cause of diarrhea worldwide. We study regulation of virulence factors in response to the surrounding environment and in this project we aim to knock out regulatory factors and virulence genes and analyze the effect on ETEC virulence. Studies on ETEC gene regulation in response to different environmental cues can help us understand disease progression and also to predict intervention therapies. The applicant will identify target genes responsible for virulence regulation and characterize these by using mutant and complemented ETEC wildtype isolates. The mutated strains will be characterized by culture and molecular studies on gene expression and also by fluorescent microscopy. The applicant will independently to construct gene deletions using molecular biology and recombinant DNA techniques appropriate for wild type pathogenic E. coli. The mutants and complemented strains will be analyzed by culture, qPCR, and fluorescence microscopy. Additional protein expression and purification are planned.
Beskrivning av kvalifikationer:
Achieved a PhD in microbiology. Documented experience in using the above mentioned methods. Additional experience of protein purification is a plus. Excellent skills in communicating both verbally and in writing and a good track record of publications in international journals are required. Ability to collaborate with a multidisciplinary group and to work independently is a merit.
