PostDoc Fellow - Ultra-sensitive detection and functional characterisation of diagnostic protein biomarkers for Alzheimer 's disease using a dynamic single-molecule readout.

At AstraZeneca, we believe in the potential of people who share our passion for science. By thinking boldly and working collaboratively to pursue discoveries beyond imagination, you'll develop further and faster than you thought possible. We'll complement your drive and determination to meet unmet patient needs around the world with the inspiration and opportunities to enhance your niche expertise and build your skills. So if you're curious about what science can do, come and make a difference.

Our Discovery Sciences team operates from truly state-of-the-art centres spanning the UK, US and Sweden. You'll work with some of the most knowledgeable technology experts in the industry, all collaborating on high-profile drug discovery projects. With a wide range of responsibilities, from sample management and screening to bio-reagent supply, the opportunities to develop your expertise here are limitless.

Main duties and responsibilities

Single molecule technologies have recently enabled the ultrasensitive detection of biomarkers to follow disease-specific processes and to monitor the effects of drug treatment. By applying total internal reflection fluorescence (TIRF) microscopy, you 'll develop novel and improve existing single molecule biomarker detection assays for Alzheimer 's disease. This will entail utilising a novel and world-unique automated TIRF platform that meets drug discovery needs.

You'll develop and implement a general concept of biomarker detection using TIRF microscopy that is independent from antibody pairs. Applying the developed method for the ultrasensitive detection of challenging biomarkers from patient samples, with a focus on neurodegenerative biomarkers, will also be part of your role. You'll demonstrate the benefit of using single molecule microscopy by comparison with traditional ELISA-based platforms and other single molecule array platforms, while establishing relationships between the variety of experimental and clinical observables to define improved biomarker strategies

You'll collaborate with academic partner Professor Fredrik Höök, Biological Physics, Chalmers University, giving you the opportunity to spend time in a stimulating academic environment.

Essential requirements
PhD and/in MD in biochemistry, biophysics or a related discipline
General expertise in TIRF microscopy
Knowledge of different surface chemistries used for optimising biosensing applications
Strong organisational and interpersonal skills
A proven record of work published in reputable scientific journals.

Desirable requirements
Application of TIRF microscopy for biochemical assays and screening using proteins, peptides and/or RNA/DNA
Wide experimental repertoire of different surface chemistry approaches for biosensing
Expertise in (biomarker/antibody) assay development
Experience in protein expression and purification
Knowledge of different biomarker detection approaches.
