The Mucin Biology Research Groups at the Sahlgrenska Academy, Institute of Biomedicine, Department Medical Biochemistry and Cell Biology has an international leading position in the research field of mucins, mucus and its protective role. Mucus is produced by goblet cells and in forms a protective layer in the intestine that separates the bacteria from epithelium. In the airways, mucus cleans and protects the epithelium by transporting bacteria and other inhaled substances upwards. The research environment is international and comprises currently about 25 researchers with additional 30 or more in collaborative groups, see also www.medkem.gu.se/mucinbiology/ . Responsible are Professor Gunnar C. Hansson and lecturer Malin E. V. Johansson. The groups have very good funding with grants from the European Research Council, National Institute of Health USA, the Swedish Research Council, the Swedish Cancer Society and others

Subject area description

The aim of the research project is to study mucus in lung and intestine with a focus on the function for protection of specific mucus proteins, as CLCA1. In the intestine the mucus layer on the epithelium is formed to keep bacteria at a distance and the dynamics around its maintenance is crucial and involves mucus secretion, processing and distal removal. This can be linked to mucus formation in the lung upon disease, as asthma, chronic obstructive pulmonary disease (COPD) and Cystic fibrosis (CF), where the mucus forms a protective layer. In healthy lungs mucus forms smaller structures that by transport clear the epithelial surface. We will study the function and regulation of CLCA1 in the intestine and its role in making intestinal like mucus in the lung in disease models of asthma, COPD and CF.

Job assignments

The work will include using a number of different methods with ex vivo analysis of mucus on intestinal or lung epithelium as the central and most important method. This is performed in a perfusion chamber using confocal microscopy or in an open system using stereo microscopy. Animal models will be used as well as human samples and cultured cells. The project will include methods in molecular biology, as RNAseq, biochemical analysis of the proteins, expression and purification of recombinant proteins and mass spectrometry for proteome studies. You are expected to be responsible for the project, but also to collaborate within the group, analyze data independently and write for publications.

Eligibility

An achieved doctoral degree is compulsory for a position as postdoctor at Göteborg University. The doctoral thesis shall be in a relevant area according to the specific position stated here. Since a position as postdoctor aims to give new holders of the doctorate the opportunity mainly to strengthen and develop their scholarly proficiency, we aim for those who have a doctoral degree not older than 3 years counting from last date of application.

Assessment

The applicant should have knowledge in intestinal protection from his/her PhD. Experience in working with animals should be supported by documentation. Experience in working with tissues in perfusion systems, confocal microscopy and mass spectrometry is strongly preferred. Experience in using a variety of methods including cell culture, molecular biology, and biochemistry is advantageous.

Employment

The employment is fulltime and temporary, two years, with placement at the Institute of Biomedicine.
