Enterotoxigenic Escherichia coli (ETEC) bacteria are the most common cause of diarrhea worldwide. ETEC are highly diverse, producing several different toxins, colonization factors (CFs) and O antigens. Our research group has developed a vaccine against such bacteria. The vaccine is based on a toxoid along with the most prevalent ETEC CFs. To evaluate if the vaccine is broad enough; i.e. protects against a majority of the clinical ETEC infections, it is of great importance to search for additional antigens (specifically CFs, but also other specific ETEC antigens) that need to be added in future ETEC vaccines. The post-doctor will identify and characterize novel ETEC CFs and other ETEC specific antigens in ETEC isolate collected from humans using "state of the art" genomic and phenotypic methods. At least 30% of ETEC bacteria causing diarrhea in humans lack a known CF. Furthermore, no ETEC specific antigens have been identified so far. Molecular studies of ETEC toxins and epidemiological studies on the prevalence of the novel ETEC antigens which will be discovered within the project will also be part of the project plan. Job assignments: Identify and characterize the above mentioned antigens in a collection of carefully selected ETEC isolates from the GU ETEC strain collection (~4000 isolates) using whole genome sequencing and advanced bioinformatic analyses. Construct mutants where the identified CF operons have been deleted for further analysis. Search for CF receptors/binding structures in different cell lines, intestinal biopsies and glyco-conjugates. Additional assignments: Animal (mouse) studies to characterize ETEC toxins and other antigens, transmission electron microscopy imaging, proteomics, transcriptomics, ETEC diagnostics (PCR and phenotypic).

Utbildning och erfarenhet
Eligibility: An achieved doctoral degree is compulsory for a position as postdoctor at Göteborg University. The doctoral thesis shall be in a relevant area according to the specific position stated here. Since a position as postdoctor aims to give new holders of the doctorate the opportunity mainly to strengthen and develop their scholarly proficiency, we aim for those who have a doctoral degree not older than 3 years counting from last date of application. The qualifications for academic positions are given in Chapter 4, Section 3 - 4 of the Higher Education Ordinance. Assessment: Achieved a PhD within 3 years. Several publications in international journals. Knowledgeable and experienced in using the above mentioned methods. Excellent skills in communicating both verbally and in writing. Regulations for the evaluation of qualifications for academic positions are given in Chapter 4, Section 3 - 4 of the Higher Education Ordinance.
