Main duties and responsibilities

PostDoc long title: Functional, biophysical and structural characterisation of a multicomponent transcription factor complex.

Nuclear hormone receptors are ligand activated transcription factors that are critical for both development and a broad range of physiological processes. We're looking for a talented postdoctoral scientist to join our Post Doc Programme to study the structure and function of a highly important full-length receptor in complex, state-of the art ligands and link this information to broader mode of action studies for active clinical projects. In addition, you'll take part in a comprehensive training programme with a focus on drug discovery and development.

You'll gain in-depth receptor experience from extensive discovery projects and technical expertise from both AstraZeneca and the academic collaboration laboratory at CBI.

Specifically, you'll design, express and purify a broad range of receptor constructs across several expression systems, as well as characterising ligands with different pharmacological profiles using biophysical techniques and functional assays. You'll also determine complex structures with either X-ray crystallography or other complementary techniques such as SAXS and high-resolution cryo-EM, and explore the allosteric pathways in between the ligand binding function and different DNA response elements and co-regulator proteins. Presenting your results at scientific conferences and writing/publishing scientific papers in high-quality, peer-reviewed journals is expected.

Essential requirements
PhD and/or MD in biochemistry, biophysics or structural biology
Significant experience of protein expression methods using mammalian and insect cells
Strong background in construct design, molecular biology and protein purification
Familiarity with biophysical and structural techniques
Genuine understanding and experience of different assay technologies
Strong organisational and interpersonal skills
A proven record of work published in reputable scientific journals.

Desirable requirements
Experience of working with the following technologies: Protein Crystallization, X-ray crystallography, Small Angle X-ray Scattering, Electron Microscopy, Isothermal Titration Calorimetry and Surface Plasmon Resonance
Experience of expressing and purifying challenging protein complexes
Experience of working with DNA binding proteins.
