Type of employment: Fixed-term employment, 4 years Extent: 100 % Location: Sahlgrenska Academy, Gothenburg First day of employment: As agreed Reference number: UR 2016/20 Job assignments: Tuberculosis (TB) is a major cause of mortality among people living with HIV. Sub-Saharan Africa bears the brunt of the HIV-fuelled TB epidemic. Co-infected patients require treatment entailing up to seven different drugs administered daily during the high-intensity two-month phase, followed by co-treatment with five drugs for another four months. Several of the agents used are known to affect the metabolism of other drugs, which may lead to sub-therapeutic drug exposure and drive resistance development. The degree of such drug-drug interactions depend on treatment regimens and the genes coding for the liver enzymes involved in the metabolic elimination of a drug in a given individual. In this project, plasma samples collected in a previously conducted clinical study in Rwandan co-infected patients will be analysed for contents of TB-drugs and their metabolites. For this purpose, a liquid chromatographic-tandem mass spectrometric (LC/MS/MS) an alytical method will be developed. The drug/metabolite concentration-time data will be analysed by pharmacometric methods, yielding models which can explain the results based on demographic, pharmacogenetic and treatment variables on the level of an individual patient. Models can be simulated to explore different scenarios (treatment alternatives, patient groups etc). It is desirable that the PhD candidate has an in-depth knowledge in pharmacokinetic theory as well as experience in mathematical modelling of biological systems, preferably with applications within the pharmaceutical sciences. Further, it is highly desirable that the candidate has theoretical and practical experience of high-performance liquid chromatographic methodology for drug quantification, especially coupled to mass spectrometry. The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral studentship.

Utbildning och erfarenhet
Eligibility: To be admitted to third-cycle studies you must fulfill both the general and the specific entry requirements. The general entry requirements is fulfilled if you have: 1. completed a degree at a second-cycle level, or 2. completed at least 240 credits worth of studies of which at least 60 credits must be at a second-cycle level, or 3. acquired equivalent knowledge in some other way in Sweden or abroad. The specific entry requirements is fulfilled if you have completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies.
