Titel: PhD student position in medical science - Mechanisms for the control of smooth muscle
Beskrivning av arbetsuppgifter:
Type of employment: Fixed-term employment, 4 years if full-time studies Extent: 100 % Location: Sahlgrenska Academy, Gothenburg First day of employment: As agreed Reference number: UR 2015/777 Job assignments: Bladder pain syndrome/Interstitial cystitis (BPS/IC) is a chronic disease, involving inflammation of the urinary bladder (cystitis). The symptoms include an increased need to urinate (increased frequency) and pain and the condition mainly affects women. Despite scientific effort there is a need to further investigate the neurophysiology, pathophysiology and pharmacology of urinary bladder inflammation. The present doctoral project will address issues regarding urinary bladder smooth muscle control and function, both in healthy and inflamed rodent bladders. By applying a broad range of techniques the functional role of purinoceptors in concert with cholinergic receptors will be investigated in control animals and during cystitis. The role of the urothelium will also be addressed and the (pato-)physiological function of the caveolae, membrane structures important for smooth muscle regulation, will be studied. The models used will involve in vitro, in situ and in vivo studies of urinary bladders from healthy rats and rats with cyclophosphamide induced cystitis as well as genetically modified knock out (KO) mice with caveolar disturbances. Furthermore, cells derived from the urothelium will be cultivated and used to validate observed results. We are looking for a PhD student, preferably with a background within the field of pharmacy, with experience from experimental work. Merits include familiarity with organbath studies (intact bladder as well as bladder strip preparations), immunohistochemistry, studies of caveolar function, cyclophosphamide treatment of rats and analysis of purines (such as ATP). The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral studentship.
