Arbetsplats: The Sahlgrenska Academy
Ort: Gothenburg
Titel: PhD student position in medical science - Phosphoinositide 3 kinases signaling in obesity
Beskrivning av arbetsuppgifter:
Type of employment: Fixed-term employment, 4 years Extent: 100 % Location: Sahlgrenska Academy, Gothenburg First day of employment: As agreed Reference number: UR 2015/1152 Job assignments - Essential Background: Obesity, a condition affecting hundreds of million people worldwide, is a leading cause for several chronic diseases and is the major cause for type-2 diabetes mellitus. A large body of evidence indicates that signal transducers at the immune-metabolism interface are implicated in the pathogenesis of obesity-associated diseases. The phosphatidylinositide 3 kinase ? (PI3K?) is a potent signal transducer, which is activated by several signals implicated in innate immunity, including chemokines and bacteria lipopolisaccarides. Importantly, PI3K? is also recruited by adrenergic signaling, which plays a potent role in several metabolic functions including lipolysis and adaptive thermogenesis. Studies from our laboratory and from our colleagues indicate that PI3K? plays a major role in the progression of diet-induced obesity, metabolic inflammation, and insulin resistance, and that PI3K? is a promising drug target for the treatment of obesity-related diseases. General and specific aims: This is a basic science research project whose general aims are: 1) Define the role of the class-1B phosphoinositide-3 kinase PI3K? in in-vivo and cell culture pre-clinical models of type-2 diabetes. 2) Investigate the role the class-1 phosphoinositide-3 kinases PI3K?, PI3Kß, and PI3K? in insulin signaling. The specific aims of this proposal are the following: 1) Investigate the role of PI3K? in the control of white adipose tissue lipolysis and browning in mice models of obesity and insulin resistance where PI3K? is ablated in specific cell types (adipocytes and neurons). 2) Investigate the role of PI3K? in pancreatic islets inflammation and ß-cell loss of function in the db/db model (C57BL/KS) of type-2 diabetes. 3) Investigate the role of different PI3K isoforms in cell culture models of insulin signaling. The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral studentship.
