Dina arbetsuppgifter
Type of employment: Fixed-term employment, 4 år Extent: 100 % Location: Sahlgrenska Academy, Gothenburg First day of employment: As agreed Reference number: UR 2015/906 Project title: Targeting mitochondria as therapeutic strategy for preventing perinatal brain injury. Job assignments: Perinatal complications, including asphyxia and preterm birth, are often associated with subsequent neurological deficits such as cerebral palsy (CP). Understanding the mechanisms triggered by insults to the immature brain will lead to the development of new strategies for the prevention and treatment of CP and other neurological deficits. Our hypothesis is that mitochondria-mediated neuronal cell death is the central process in perinatal brain injury. The current project will focus on the role of the mitochondria related protein, apoptosis inducing factor (AIF), in perinatal brain injury. The project includes several distinct but closely related aims that will study different aspects of mitochondria functions in relationship with brain injury. The focus will be on the impact of regulation mitochondrial metabolism on neonatal brain injury in animal models, the role of inflammation and autophagy on mitochondria dysfunction and brain injury, as well as the role of AIF release in neuronal cell death and brain injury. We will apply conditional gene deletion approaches to assess the role of AIF in perinatal brain injury. Furthermore, a unique gene knock-out strategy will be used to modulate AIF release and thereby acquire specific information on the importance of AIF release from mitochondria to the cytosol. Finally, the neuroprotective efficacy of a peptide will be evaluated which prevents AIF translocation to the nucleus, wher e it induces DNA degradation. The project will help to gain new insights for understanding the pathogenesis of perinatal brain injury and lead to the discovery of new prevention / neurprotective strategies. We would like the applicants to motivate why they interested in the project and why they are suitable applicants. The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral studentship.
