Type of employment: Fixed-term employment, 4 years Extent: 100 % Location: Sahlgrenska Academy, Gothenburg First day of employment: As agreed Reference number: UR 2015/1098 Job assignments: Chronic hepatitis B virus (HBV) infections may cause severe complications such as hepatocellular carcinoma and liver cirrhosis. Patients who need long-term antiviral treatment to prevent these complications are identified by means of HBV DNA and surface antigen (HBsAg) quantification in serum. Therefore it is important to clarify mechanisms that explain these serum levels, including immune clearance of infected liver cells and free virus, host suppression of viral replication, and differences between viral strains that influence the production of virions or the subviral particles (SVP) that constitute most of the HBsAg in serum. This project is a continuation of our previous work, and comprises analyses of liver biopsies and in vitro studies using a cell line that expresses the recently discovered HBV receptor. By analysing liver biopsies using immunohistochemistry staining we will study how levels of HBV DNA and HBsAg in serum reflect the proportion of infected hepatocytes, and by FISH staining of HBV genomes we will study the distribution and spread of wild-type and variant strains of HBV. Liver biopsies from patients whose clinical stage and infection characteristics have been previously investigated in detail, will be analysed. HBV infection of HepG2 cells that express the HBV receptor NTCP will be performed in order to study the impact of HBV genotype and mutations on viral infection and replication. Standardised doses of HBV from patients, representing genotypes A-D, with or without certain mutations will be compared. The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral studentship.

Beskrivning av kvalifikationer:
Eligibility: To be admitted to third-cycle studies you must fulfill both the general and the specific entry requirements. The general entry requirements is fulfilled if you have: 1. completed a degree at a second-cycle level, or 2. completed at least 240 credits worth of studies of which at least 60 credits must be at a second-cycle level, or 3. acquired equivalent knowledge in some other way in Sweden or abroad. The specific entry requirements is fulfilled if you have completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies.
