The Sahlgrenska Academy, Gothenburg

PhD student position in medical science - BMP4, novel treatment target for obesity and T2DM

Type of employment: Fixed-term employment, 4 years Extent: 100 % Location: Sahlgrenska Academy, Gothenburg First day of employment: As agreed Reference number: UR 2015/909 Project title: Regulation of white and beige/brown subcutaneous adipogenesis - role of BMP4 and its secreted inhibitors Gremlin1 and Noggin Job assignments: The global epidemic of obesity is the major cause of the dramatic increase in Type 2 diabetes. Obesity promotes insulin resistance, an important pathogenic factor for the development of Type 2 diabetes as well as other chronic diseases including cardiovascular disease. However, several studies have shown that fat mass per se is not the direct cause, but rather the associated dysregulated and inflamed adipose tissue accompanying hypertrophic obesity. Several recent studies have shown that adipose precursor cells in the subcutaneous adipose tissue are multi-functional and can assume either a classical white lipid-storing phenotype or a more beneficial, oxidative beige/brown phenotype. There is current much global interest in understanding how this is regulated and, even more, if this can be induced in man as a potential therapeutic target in the treatment of obesity and Type 2 diabetes. Our recent work has identified BMP4 and its inhibitors Gremlin 1 and Noggin as key regulators of the development of adipogenic precursor cells into a white or a brown/beige phenotype. However, the molecular mechanisms involved need to be characterized in detail. The objectives of the present proposal will allow the PhD student to participate in a large, but well-focused, project aimed at understanding the mechanisms whereby BMP4 and its inhibitors regulate adipose tissue and the associated in vivo consequences. The project will include basic molecular methods using both established cell lines and primary human adipose precursor cells to elucidate the molecular mechanisms. In addition, an important part of the project will be to characterize the appropriate animal models generated in the project and the associated metabolic phenotype. We would like the applicants to motivate why they interested in the project and why they are suitable applicants. The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral studentship.

To be admitted to third-cycle studies you must fulfill both the general and the specific entry requirements. The general entry requirements is fulfilled if you have: 1. completed a degree at a second-cycle level, or 2. completed at least 240 credits worth of studies of which at least 60 credits must be at a second-cycle level, or 3. acquired equivalent knowledge in some other way in Sweden or abroad. The specific entry requirements is fulfilled if you have completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies.
