Arbetsplats: The Sahlgrenska Academy
Ort: Gothenburg
Titel: PhD student position in medical science - Tissue-engineering of whole pancreas
Beskrivning av arbetsuppgifter:
Type of employment: Fixed-term employment, 4 years Extent: 100 % Location: Sahlgrenska Academy, Gothenburg First day of employment: As agreed Reference number: UR 2015/961 Job assignments: Diabetes mellitus is a global disease with immense economic and social burden. However, the treatment of diabetes mellitus remains inadequate. Regenerative medicine approaches such as a bioengineered pancreas may be a potential therapeutic alternative. A bioengineered pancreas will benefit from the development of a bioscaffold that supports and enhances cellular function and tissue development. Perfusion-decellularized organs are a likely candidate for use in such scaffolds since they mimic compositional, architectural and biomechanical nature of a native organ. Decellularization is the process of removing cells/DNA using physical, chemical or enzymatic factors. In this project, we will investigate perfusion-decellularization of whole pancreas and the feasibility to recellularize the whole pancreas scaffold with stem cells. The acellular scaffold will be characterized thoroughly for the various ECM components, and tested for DNA, biomechanical properties and growth factor determination. The scaffold will be seeded with human stem cells and tested by molecular techniques for differentiation to insulin-producing beta cells. These studies will advance the field of regenerative medicine towards the development of a bioengineered pancreas capable of supporting and enhancing pancreatic cell functionality and may in the long term be used for transplantation to cure diabetes mellitus. The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral studentship.
