Reference number: UR 2016/237. Project title: Molecular defects in the protective mucus in colitis. Job assignments:The intestinal tract is a harsh milieu with chemical, physical and microbial hazards. The bacterial load in colon is an extreme challenge and the outermost defense is a physical barrier of densely packed and well-organized mucus constantly renewed by secretion from the mucin producing goblet cells. This creates a zone restricting bacteria to have contact with the epithelium. A defect mucus barrier leads to massive bacterial contact with the epithelium causing colitis and cancer development both in animal models and in Ulcerative colitis patients. The project aims to understand the molecular details causing mucus defects that lead to colitis and cancer. The PhD student will study ion transport and its effect on mucus layer formation, mucus secretion from goblet cells, the mucus and epithelial protein composition and mucus turnover. The student will work with cell systems, animal models and human samples and be able to learn different methods as: Mucus measurements ex vivo with interventions, mucus quality analysis with confocal microscopy, electrophysiology, proteomics using mass spectrometry, expression analysis, immunohistochemistry, in vivo labeling to analyze turnover and other cell biology, molecular biology and biochemistry methods. The work will be performed in one of the Mucin Biology groups and the environment is recognized as world leading in this field. The Mucin Biology groups have together a broad methodological expertise which will be available for the PhD student's training and guidance. The milieu is international with English as the working language, as the group consists of students and postdocs from different countries and there are several international collaborators. A better molecular understanding of defects in protection of the epithelium and defects causing disease is important to develop better treatment strategies and to develop new therapies that can improve the protective mucus barrier.

Utbildning och erfarenhet
Eligibility: To be admitted to third-cycle studies you must fulfill both the general and the specific entry requirements. The general entry requirements is fulfilled if you have: 1. completed a degree at a second-cycle level, or 2. completed at least 240 credits worth of studies of which at least 60 credits must be at a second-cycle level, or 3. acquired equivalent knowledge in some other way in Sweden or abroad. The specific entry requirements is fulfilled if you have completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies.
