Project title
Herpes virus infections in the CNS -genetics, pathogenesis and antiviral treatment
Job assignments
This project concerns the pathogenesis of clinically important infections with neurotropic herpesviruses, and combines experimental and clinical virology. The aim of the project is to elucidate pathogenetic mechanisms trough analysis of viral genetics and correlation of these results with clinical data and viral load during CNS-infections caused by herpes simplex virus (HSV)-1, HSV-2 och varicella-zoster virus (VZV) in children and grownups with encephalitis/meningitis/myelitis or vasculitis-induced stroke. The work includes PCR-based sequencing (in-house methods) and deep sequencing (international collaboration and transfer to in-house methods). These analyses are performed directly on patient samples of cerebrospinal fluid (CSF).
The bioinformatic part focuses on genetic diversity within the population and the patient, including detection of different genotypes and recombinants thereof. In addition, viruses carrying virulence markers and ge netic variants carrying resistance mutations towards the antiviral compound acyclovir will be sought for. The result from viral sequencing and analyses thereof may then be correlated to clinical data concerning sequels, mortality and prognosis. Furthermore, determination of acyclovir concentrations will be performed and correlated to results of viral genetics. The project includes functional studies of viral envelope glycoproteins of HSV-1/HSV-2/VZV through cell culture experiments evaluated by molecular, chemical and biochemical methods and by confocal microscopy using fluorescence-labeled viruses/virus mutants. The results may be of importance for treatment and prophylaxis of infections with neurotropic herpes viruses.
PhD positions are financed mainly through education grants during the first two years and employment as doctoral students during the last two years and pertains to a doctoral degree.
Eligibility
The general entry requirements are fulfilled if you have either 1, a second cycle degree (usually described as a masters degree) or 2, completed courses at an academic level corresponding to at least 240 education credits. At least 60 of these credits must be at second cycle level. 60 credits is equal to one year of full-time studies. You also need to fulfill the specific entry requirements which is an English language requirement; Engelska B.
Assessment
After the application deadline has passed, an initial assessment of the candidates eligibility is made at the faculty office and sent to the project supervisor for selection. The process can then take up to 6 months before a candidate is selected.
