Examensarbete höstterminen 2014, 30 hp

Title:

"Transport and mobility in and release from extended release amorphous systems"

AIM: Investigate the link between mobility and structure (plasticizing effect) in the dry state or almost dry state to the aqueous phase behaviour and release properties for hot melt extruded solid dispersions based on model drugs and extended release matrix formers.

OUTLINE: Prepare solid dispersion (melt and solvent methods) using celullose and polyethylene oxide based carriers and use model drugd such as ibuprofen and felodipine. Look at the effect of history induced during manufacturing. Probe the mobility and structure of the dry state and the effect of humidity (max 2 week storage). Determine dissolution characteristics as well as phase behavior of dissolved material. Compare behavior for system with and without model drug. Keep drug load relatively low (about 20%). Potentially investigate the effect of salting-out ions in the composition.

METHODS:

1. Use HME extrusion to manufacture zero porosity pellets or films.

2. Investigate the mobility of the model drug substance dielectric relaxation spectroscopy (and potentially calorimetric methods).

3. Investigate the structure and phase behavior of extruded material using modulated DSC, XRD & Raman.

4. Investigate phase behavior in solution using transmittance (cloud point) measurements.

5. Investigate release behavior using USPII or similar.

6. Investigate physical stability under stressed conditions and means to probe early changes.
