Immune evasion in gastrointestinal adenocarcinomas by adenosine from CD39+ Treg

Description of the research project
The immune system has the ability to recognize and kill tumor cells. Cytotoxic and cytokine producing T cells, as well as natural killer (NK) cells, are important for anti-tumor immunity, but in order to be effective, they need to be present within the tumor. However, so called regulatory T cells (Treg) inhibit the function of conventional activated T cells and NK cells, and may thus help the tumor to avoid attack from the immune system. We have recently shown that Treg from cancer patients inhibit transendothelial migration of conventional T cells, thereby probably preventing anti-tumor immunity through a unique mechanism by preventing T cell accumulation in the tumor. In this project, we will investigate Treg expression and activity of the ectonucleotidase CD39, that reduces ATP to adenosine. We will also determine how release of adenosine affects endothelial cells and inhibits recruitment of activated T cells.
The major part of the project will be carried out in collaboration with clinical researchers, using patient material for isolation of lymphocytes and functional in vitro studies. The methods to be used include flow cytometry for both characterization and sorting of cells, immunohistochemistry, PCR, and an in vitro model of lymphocyte transendothelial migration. We will also use a mouse model of spontaneous colon adenocarcinoma for in vivo studies of Treg activity. We hope that these studies will elucidate the mechanisms preventing recruitment of effector lymphocytes to tumors, and define novel mechanisms of Treg activity on endothelial cells, a completely new target for Treg. The project may therefore be important in the development of improved immunotherapies for cancer patients based on CD39 inhibition.
