Nuclear transcription - the Mediator complex

Description of the research project
Signal transduction pathways trigger a wide variety of physiological responses, one of the most notable of these being changes in Pol II activity via the multiprotein Mediator complex. Mediator is essential for basal and regulated expression of nearly all Pol II dependent genes and depletion of human Mediator from nuclear extracts abolishes Pol II transcription. Gene specific activator proteins stimulate transcription by recruitment of general transcription factors (GTFs) to promoters. In this process, Mediator supposedly functions as a bridge between the regulatory proteins and the basal pol II transcription machinery. Direct interactions have been demonstrated between many activators and specific Mediator subunits, e.g. -catenin, p53, Gli3, and nuclear hormone receptors.
In the suggested project, we will investigate the role of Mediator in cell cycle dependent regulation of transcription. As a model system, we will use fission yeast (Schizosaccharomyces pombe), but we also intend to analyze the impact of Mediator on cell cycle dependent transcription in mammalian cells.
Our group uses a wide range of techniques in the laboratory, ranging from structural biochemistry to mouse genetics. Currently, the group consists of one senior scientist, three postdocs, and four PhD students. In addition to work with nuclear transcription, the group also studies mitochondrial gene expression. Our laboratories are located in the Department of Medical Biochemistry and Cell Biology, Medicinaregatan 9A. The research group has a strong international character and the working language is English.
Desired background
Suitable candidates should be self-motivated and have an intense interest in research. The planned research project will be performed very independently and the work will at times be extremely challenging. Applicants may have a background in biochemistry or molecular biology, and preferably also experience in fission yeast genetics.
