Transplantation of induced pluripotent stem cells after radiotherapy

Description of the research project
Aims: We aim to find treatment strategies that ameliorate the severe late effects seen after radiotherapy to the young brain. We create induced pluripotent stem cells (iPSCs) through transient, non-viral transfection of mouse and rat fibroblasts and transplant them into the hippocampus of irradiated young rats and mice.
Background: A single, moderate dose of IR causes a life-long depletion of stem cells in the hippocampus, an area important for memory and learning. We found that physical exercise ameliorated the injury.
Preliminary results: Transplantation of stem cells into the hippocampus of irradiated mice also ameliorated the injury and improved memory function. These stem cells were from a cell line, they were not iPSCs. Very few labs in the world so far can create iPSCs through transient, non-viral transfection. The lab of Carsten Culmsee, Marburg, Germany is one of them, and we have an established collaboration since many years. Transient transfection minimizes the risk of teratoma induction or even malignant transformation due to viral gene insertion.
Work plan: Mouse fibroblasts have been transformed into iPSCs and will be used for transplantation. Rat iPSCs will be created. Transplanted rodents will be subjected to extensive functional testing.
Methods: We have established a model for irradiation-induced injury in rats and mice. We have established protocols for transplantation of cells to mouse brains. For functional testing studies we employ the novel IntelliCage system and other tests of motor function and cognition.
Significance for childhood cancer: If we can mitigate the adverse late effects of radiotherapy in the increasing number of survivors of childhood cancer, we could greatly improve their quality of life. Successful use of iPSCs means that in the future, cells from the patient can be transformed to stem cells and used for transplantation.
