Doktorandplatsen avser två års utbildningsbidrag och två års doktorandanställning, alternativt fyra års doktorandanställning, och beräknas leda fram till doktorsexamen.

Pharmacokinetics and metabolism of the antimalarial drug piperaquine
Description of the research project
Preliminary in vitro enzyme inhibition data suggest that piperaquine and some of its urinary metabolites are potentially potent inhibitors of major drug metabolizing Cytochrome P450 enzymes. An inhibition of one or more of these enzymes could lead to clinically important drug-drug-interactions. The inhibitory effect of piperaquine could be especially remarkable since it could potentially persist several months after treatment due to its long terminal half-life of about one month.
Project activities:
1. Develop a liquid chromatographic - mass spectrometric (LC-MS/MS) method for quantification of piperaquine and its metabolites in human plasma.
2. In vitro investigations of the inhibitory effects of piperaquine and metabolites on drug metabolizing Cytochrome P450 enzyme activities.
3. Bio- and dataanalysis of clinical piperaquine study in Vietnamese healthy subjects with the aim to determine the terminal half-life of piperaquine and metabolites and to investigate dose-linearity. Quantification in plasma and kinetic evaluation of the putative CYP3A4 probe 4?-hydroxycholesterol.
4. Design and conduct of a directed clinical inhibition study exploring the inhibitory effects of piperaquine and/or metabolites on CYP activity using conventional and validated enzyme-specific probes.
Desired background
The candidate should have a solid background in pharmacokinetics and drug metabolism. A basic degree in the pharmaceutical sciences is desired as well as some previous experience with drug quantitation by LC-MS/MS.
