Development of reliable experimental models for stress-induced cardiomyopathy. Pathophysiology and treatment of stress-induced cardiomyopathy.

Description of the research project
The aims of this project are: 1) to establish relevant small animal models of stress-induced cardiomyopathy (SIC), 2) to decipher the pathophysiology behind SIC and 3) to develop a pharmacological treatment strategy.
Using a state of the art high resolution ultrasound system specifically designed for non invasive imaging of small animals, we will set up mouse and rat models of SIC. The cardiac function will then be quantified by echocardiography as well as by advanced invasive hemodynamic and electrophysiological techniques.
The most favored hypothesis regarding the pathogenesis of SIC is a stress-induced catecholamine mediated cardiotoxic effect. Our preliminary results from animal models directly support the catecholamine-toxicity hypothesis, as SIC-like cardiac dysfunction develops in both rats and mice in response to catecholamine stress.
We will test the hypothesis that lipotoxicity is responsible for the cardiac dysfunction observed in SIC. We will study genetically modified mice to delineate the role of androgens and estrogens in SIC. Adenosine and other pharmacological treatments will be tested in these models.
Knowledge of mechanism involved in the development of SIC is important because this disease is becoming increasingly recognized, little is known about its pathogensis and no specific treatment exists.
