The research area and duties
The project is funded by the Swedish Research Council (2020-2024) and involves researchers from Linnaeus University, Lund University and The McGill University, Montreal, Canada. The overall aim of the lab in Kalmar is to deepen our understanding of the evolutionary optimized contractile function of muscle and gain insight into how changes in muscle proteins cause deterioration in diseases and in ageing. On-going projects also aim to exploit muscle proteins in nanotechnological applications. The specific aim of the present PhD project will be to gain in depth insights into the molecular basis for high resistance to stretch of active muscle (skeletal muscle and heart) with low consumption of chemical energy (ATP). In addition, diseases causing genetic mutation of muscle proteins will be studied. In the focus will be hypertrophic cardiomyopathy the leading cause of sudden cardiac death in young people. The studies will be performed using unique methodology in a lab team of a senior scientist and a postdoc in addition to the main supervisor. The methodology expected to be central in the PhD studies includes:
i. a novel virus-free expression and purification system for the molecular motor myosin II of human skeletal and cardiac muscle
ii. recording of force and ATP turnover from small groups of isolated myosin molecules when length changes, corresponding to stretch of a muscle, are imposed.

Entry requirements
A candidate fulfils the general entry requirements (as laid out in the Swedish Higher Education Ordinance (Högskoleförordningen), Chapter 5 and 7) if he/she:

• Has been awarded a degree on second-cycle level
• Has completed courses totalling at least 240 higher education credits (ETCS), of which at least 60 must be at second-cycle level in a related field, or acquisition of essentially equivalent knowledge in Sweden or abroad.

Assessment grounds
Other assessment criteria are:

• A Master of Sciences degree or equivalent in a subject area of relevance for the project (chemistry, biology, biomedicine, biotechnology etc.)
• High proficiency in written and spoken English
• Experience and skills in fluorescence microscopy, cell cultivation and gene technology as well as protein expression, purification and characterization
• Important personal qualities are accuracy, creativity, good problem-solving skills, flexibility, independency but also capability to work in teams

The selection of candidates is made with regard to the applicants' ability to successfully complete and benefit from their studies at the graduate level.

Terms of employment:
The vacancy is a fully funded full-time four years appointment with the objective to obtain a PhD, starting as soon as possible after application deadline. The employment as a doctoral student is limited according to chapter 5, § 7 of the Higher Education Ordinance. The total employment period may not be longer than the equivalent full-time postgraduate studies of four years. Cooperation with other research groups is part of the research tasks and a stay at another university/company may occur.
