Description of task:
This Diploma work aims to explore the role of lactose fines in dry powders for inhalation with specific focus on formulations including a coating agent. The main questions to be addressed are:
Do lactose fines behave in the same way as the API?
How sensitive are the lactose fines to processing conditions?
Is there a way to correct for the lactose fines in laser diffraction methods of aerosolization behavior?
To address these questions, a series of formulations will be produced using high shear mixing. Special focus will be on the influence of mixing time. The formulation will be filled into a prototype device and analyzed using the standard technique, i.e. NGI impactor analysis. Both the distribution of API and of lactose in the aerosol will be investigated.
The formulations will also be analyzed using the Sympatec Helos particle sizer which is based on laser light diffraction. Different set ups will be used, to capture both the full particle size distribution, and the particle size distribution of the aerosol at inhalation conditions.
Data analysis will focus on the differences observed between API and lactose fines, and correlations between NGI data and Sympatec data with regard to the three questions above.
Relevant education for this work is believed to be:
Chemical engineering, Chemistry, Physics, Technical physics or Pharmaceutical studies with some knowledge within pharmaceutics, particle sizing and aerosol science.
