Our New Modalities (NMs) Chemistry team in Respiratory, Inflammation & Autoimmunity leads the efforts focused on rapidly growing the use of novel peptide and oligonucleotide-based modalities and molecular conjugates in the treatment of respiratory diseases. We achieve this in close collaboration with excellent scientists from interdisciplinary departments - all joined by the goal to discover the respiratory medicines of the future.

We offer a possibility for you to be part of the computational chemist team as an expert within Oligonucleotides Design. You and your colleagues are specialized on design of both SMs and NMs based therapeutics. As an authority in the field, you will contribute to the design of new oligonucleotide sequences to specific RIA projects.

Main Duties and Responsibilities

We expect you to design novel oligonucleotides and to influence the selection of portfolio targets by highlighting opportunities for therapeutic applications. You will be an active member of the RIA oligonucleotide projects from target selection to identification of clinical drug candidates. Working and navigating in our multidisciplinary environment as well as mentoring junior colleagues will give you the possibility to practice and develop your leadership skills.

* Lead oligonucleotide design in individual project teams
* Demonstrate state of the art oligonucleotide design options for both target validation and therapeutic use
* Analysis of mRNA/microRNA target sites for ASOs/siRNAs/antimiRs
* Identification of suitable mRNA sequence regions for RNAi (UTRs, coding regions)
* Identification and correlation of sequence and potency parameters for optimization
* Project specific display of sequence alignments and potency data
* Contribution to internal projects and collaborations with external partner organizations
* Drive our scientific reputation by publications as well as giving presentations internally and externally and in collaboration with external organizations

Essential Requirements

Your success in this role will depend of your ability to combine your deep scientific knowledge with excellent interpersonal skills. In addition, you have the ability to engage and collaborate across boundaries with a can-do attitude and a clear delivery focus.

* PhD in bioorganic chemistry, computational chemistry or bioinformatics
* Experience in pharmaceutical drug discovery of oligonucleotide drug candidates
* Validated in-depth knowledge of recent developments in oligonucleotide drug discovery
* Robust understanding of mechanism of action of oligonucleotide therapeutics (alternative splicing, steric blocking of protein translation, RNAaseH activity, RISC loading, splice switching, immune stimulation)
* Expertise in conformational/secondary/tertiary structure analysis of mRNA using state of the art software
* Expert user of ASO modeling software for e.g. prediction of ASO-target binding energies
* Experience in molecular dynamics simulations, preferably for NMs
* Basic knowledge in prediction of potential off-target interaction and toxicity
