Our Mission
AstraZeneca is committed to the development of the next generation innovative respiratory medicines. The New Modalities (NMs) Chemistry team in RIA leads the efforts focused on expanding the use of novel peptide and oligonucleotide-based modalities and molecular conjugates in the treatment of respiratory diseases. The New Modalities team achieves this in close collaboration with computational chemists as well as scientists in the Bioscience, Translational Science & Bioinformatics, DMPK and Pharmaceutical Sciences departments - all joined by the goal to discover the respiratory medicines of the future.
As an Associate Principal Scientist/Principal Scientist in the Computational Chemistry team, you are surrounded by computational chemists specialized in the design of both SMs and NMs based therapeutics. As a RIA authority in the field of oligonucleotide design, you contribute as an active project team member to the selection and design of new oligonucleotides to specific RIA projects, in close collaboration with colleagues in other departments. In addition, you will influence the selection of portfolio targets by highlighting opportunities for therapeutic application of oligonucleotides. Overall, you will be part of RIA oligonucleotide projects from target selection to identification of clinical candidates.
Main Duties and Responsibilities
We expect you to have excellent interpersonal skills necessary for working in our multidisciplinary environment and to act as mentor for more junior colleagues.
Furthermore, you have an excellent scientific background with strong written and oral communication skills supplemented by an ability to engage and collaborate across boundaries with a proactive attitude and a clear delivery focus.
You will get the opportunity to:
* Lead oligonucleotide/sequence design in individual project teams as active project team member
* Actively impact RIA 's target portfolio by suggesting options for oligonucleotides for both target validation and therapeutic use, leveraging state of the art oligonucleotide design
* Collaborate seamlessly with RIA 's bioinformatics experts
* Contribute to internal projects and collaborations with external partner organizations
* Analysis of mRNA/microRNA target sites for ASOs/siRNAs/antimiRs to support generation of RNAi oligonucleotide libraries
* Identify suitable mRNA sequence regions for RNAi (UTRs, coding regions)
* Identify and correlate sequence and potency parameters for optimization
* Project specific display of sequence alignments and potency data
* Drive RIA IMED scientific reputation by identifying and progressing publication opportunities internally, across AZ and/or together with external partners
* Contribute to conference and project presentations internally and externally
Essential Requirements
* PhD in bioorganic, bioinformatics or computational chemistry
* In-depth knowledge of recent developments in oligonucleotide drug discovery
* Robust understanding of mechanism of action of oligonucleotide therapeutics (alternative splicing, steric blocking of protein translation, RNAaseH activity, RISC loading, splice switching, immune stimulation)
* Expertise in conformational/secondary/tertiary structure analysis of mRNA
* Expert user of ASO modeling software for e.g. prediction of ASO-target binding energies
* Basic knowledge in prediction of potential off-target interaction and toxicity
Desirable Requirements
* PostDoc experience and professional experience in drug discovery of oligonucleotide drug candidates
