1 Research student (2 years) in the research education subject: structural biology
study the structure and assembly mechanism of surface virulence organelles in Gram-negative pathogens
Most Gram-negative pathogens express adhesive surface organelles that allow them to specifically target and bind to host tissues and cause infection. The biogenesis of such organelles is a complex and challenging task that involves secretion of organelle subunits through the inner and outer membranes, their transport through the periplasmic space, subunit folding and fiber assembly. The chaperone/usher (CU) machinery assists in biogenesis of the most ubiquitous and versatile virulence organelles. CU assembled adhesive organelles are not only important virulence factors and promising drug targets, but many of them are also highly efficient protective antigens suitable for developing novel subunit vaccines against different infectious diseases. This PhD project will be centered on elucidation of structure, assembly mechanism, and function of several CU assembled poly-adhesive organelles from pathogenic strains of Escherichia, Salmonella, and Yersinia. The PhD student will also participate in the developing a high throughput procedure to identify novel immunoprotective antigens, potential candidates for subunit vaccines against infectious diseases. The research methods will include gene cloning and design, protein expression/co-expression, recombinant protein production, X-ray crystallography, electron microscopy, and various immunological assays.
Qualifications
Applications are welcome from students preferably with experience in biochemistry, genetic engineering, microbiology, molecular biology or related fields. Experience in methods of protein chemistry, expression vectors design, X-ray crystallography, and immunology is a significant advantage
