Subject area

Immunoregulation in intestinal tumors.

Subject area description

The overarching goal of our research is to determine mechanisms of immunoregulation in autoimmune diseases and tumor immunity. In this project we study immunoregulation in a mouse model for human coloncancer (the APCmin/+ mouse) that spontaneously develops tumors in the intestine. We have recently shown that natural killer T lymphocytes (NKT cells) have an immunoregulatory function in intestinal tumors, promoting tumor growth. By enhancing regulatory T lymphocytes and inducing an immunosuppressive environment in the tumor, an efficient immune response against the tumor is prevented. Moreover, tumor NKT cells are functionally altered and have a different transcription factor setup compared to NKT cells in lymphoid tissues. The aim of the announced project is to determine how the tumor affects the NKT cell functional capacity, and to identify the underlying mechanisms by which NKT cells promote an immunosuppresive environment.

Job assignments

Mechanistic studies of the immunoregulatory function of NKT cells in the APCmin/+ model using different genetically modified mice, cell sorting and transfers, flow cytometry, immunofluoresence and a diversity of molecular biology and cellular immunological methods. Total mRNA analysis and bioinformatics analysis are also included. The project also encompasses typing and responsibility for breeding and crossing of strains of genetically modified mice.

Eligibility

An achieved doctoral degree is compulsory for a position as postdoctor at the University of Gothenburg. The doctoral thesis shall be in a relevant area according to the specific position stated here. Since a position as postdoctor aims to give new holders of the doctorate the opportunity mainly to strengthen and develop their scholarly proficiency, we aim for those who have a doctoral degree not older than 3 years counting from last date of application.

The qualifications for academic positions are given in Chapter 4, Section 3 - 4 of the Higher Education Ordinance.

Assessment

We are looking for a motivated and enthusiastic postdoc that is able to work independently. A strong background in basic immunological research, and extensive experience with mouse work, murine disease models, breeding and typing is a requirement. Extensive experience with flow cytometry sorting and analysis is also required. Knowledge and research experience on T lymphocytes, immunoregulation, tumor immunology, and a broad methodological competence, including molecular biology, is a merit.
