Type of employment: Fixed-term employment, 4 years Extent: 100 % Location: Sahlgrenska Academy, Gothenburg First day of employment: As agreed Reference number: UR 2015/907 Project title: Targeting energy metabolism in macrophages to promote plaque regression in atherosclerosis Job assignments: Atherosclerosis is one of the main causes of death and disability in the world. In a majority of cases, atherosclerosis development proceeds sub-clinically until the patient suddenly experiences a sudden myocardial infarction or stroke. At this stage, advanced lesions are already present. Thus it is pivotal to find treatment strategies to heal advanced atherosclerosis. Inflammation within atherosclerotic lesions consumes large amounts of oxygen and glucose. As a consequence, hypoxia and glucose depletion develops deep in advanced lesions. In these areas, macrophages cannot maintain adequate ATP production as shown in animal models of atherosclerosis. ATP depletion impairs the ability of macrophages to remove lipid from lesions. The first aim of this PhD project is to investigate if advanced human atherosclerotic lesions also develop local hypoglycemia and ATP depletion. The second Aim is to use cell culture and animal models to address the hypothesis that restoration of macrophage ATP levels may help heal advanced lesions. Our hypothesis is that if the intense inflammation is reduced, by manipulation of T-cell receptors in advanced lesions, oxygen and glucose availability will increase in the core thus restoring macrophage ATP levels. Restored ATP levels make it possible for macrophages to remove lipids from the plaque. This may constitute a principally new, clinically feasible, approach to treat advanced atherosclerosis. This doctoral thesis will be performed at the Wallenberg laboratory ( http://www.wlab.gu.se/ ), and will use a translational approach to explore the importance of plaque energy metabolism on atherosclerosis progression and regression. This includes working with human plaques, atherosclerotic mouse models, advanced imaging techniques and dynamic three dimensional cell culture studies. We would like the applicants to motivate why they interested in the project and why they are suitable applicants. The PhD position comprises 4 years full-time studies and leads to a doctoral degree. The position is funded via doctoral studentship.

Utbildning och erfarenhet
To be admitted to third-cycle studies you must fulfill both the general and the specific entry requirements. The general entry requirements is fulfilled if you have: 1. completed a degree at a second-cycle level, or 2. completed at least 240 credits worth of studies of which at least 60 credits must be at a second-cycle level, or 3. acquired equivalent knowledge in some other way in Sweden or abroad. The specific entry requirements is fulfilled if you have completed the English B/6 course or is considered to have acquired equivalent knowledge through previous studies.
