Project title:
Cortical porosity - its regulation and association with fracture

Job assignments
Osteoporosis (bone fragility) is a large public health concern in Sweden today and causes approximately 70.000 fragility fractures yearly. The mortality the first year following hip fracture is 20% for women and 30% for men. In the elderly, most fractures occur at mainly cortical bone sites, such as the hip, humerus and forearm. Experimental studies have demonstrated that cortical porosity is a strong determinant of bone frailty but little is known of its role in predicting fractures in the elderly.

The primary aim of the proposed project is to determine if cortical porosity, measured using High Resolution peripheral Quantitative Computed Tomography, is higher in well characterized elderly fracture patients than in age matched controls. The secondary aims of the project are to increase the knowledge of the environmental regulation of cortical porosity. The association between cortical porosity and various types (with different loading) and dur ation of physical activity, dietary intake (especially intake of calcium and vitamin D), and smoking will be investigate in young and elderly subjects. Serum markers of signaling proteins and hormones (e.g. sclerostin and parathyroid hormone perceived to be involved in the skeletal response to mechanical loading) will be measured to investigate whether associations found are mediated by these markers. Increased knowledge in the regulation of cortical porosity and its ability to predict fracture could lead improved fracture prevention.

PhD positions are financed mainly through education grants during the first two years and employment as doctoral students during the last two years and pertains to a doctoral degree.

Eligibility
