Position

We invite applications for a three-year Early Stage Researcher (Ph.D. student) position being part of the Marie Sklodowska Curie Initial Training Network transMed. The young researcher will be involved in a multi-disciplinary drug development project focusing on a novel class of compounds with strong neuroprotective properties in diseases of the retina.

The position will be a three year employment contract at RISE in Södertälje in Sweden. The goal is to reach the PhD title. The candidate will be enrolled at the faculty of pharmaceutical sciences at the University of Iceland that will give the degree. The University of Iceland is a full partner in the Marie Sklodowska Curie Initial Training Network transMed.

Subject

Neurodegenerative diseases of the retina are a major cause of blindness and are often untreatable. The project concerns the development of lipid formulations such as liposomes and solid lipid nanoparticles that enable delivery of the novel cyclic nucleotide analogues to the retinal tissue in the eye. Formulation development will build on previous work using glutathione and polyethylene glycol coated vesicles to target specific endogenous transport systems in the blood-retinal barrier. Focus of the present thesis will to optimize the delivery to the retina using different types of lipid systems and the adaptation of such vehicles for large scale, aseptic or sterile manufacture that will allow the use of the drug delivery systems in clinical studies. The vesicles will be optimised based on efficacy of drug delivery to the target tissue, loading efficiency, scalability, manufacturing process, environmental impact and cost. The different types of liposomes and solid lipid nanoparticles will be tested using established in vitro and in vivo models for retinal disease. It will pave the way for further development for every day therapeutic use in patients.

Job description

The methods used in during the thesis will include formulation and preformulation work, set up of dissolution, and in-vitro permeability methods, micro fluidizing techniques, analytical techniques such as liquid chromatography, characterization techniques such as small angle X-ray scattering, confocal scanning microscopy, SEM and TEM and particle size measurements such as dynamic laser light scattering. Preclinical experiments to study the in vitro and in vivo effects of the drug delivery systems will also be included in the thesis work.

The critical requirements for the drug delivery systems will be identified and the design space of the vesicles will be clarified. Manufacture of formulations will be done at a scale and with a process suitable for clinical studies. The project will be performed in a strong international collaboration with partners from academia and industry, located in six different European countries

Eligibility criteria

The Early Stage Researcher candidate









* shall not have resided and not have carried out his/her main activity (work, studies, etc.) in Sweden for more than 12 months in the 3 years immediately prior to the recruitment date
* shall be in the first four years (full-time equivalent research experience) of his/her research career
* shall have a degree allowing him/her to start a PhD thesis in Sweden
* shall not have been awarded a doctoral degree

Skills/Qualifications

We are searching for an Early Stage Researcher / PhD student who is interested and should have some experience in at least one of the following fields: Pharmaceutics, Biology, Physical chemistry, Biotechnology. One to four years of research experience in Pharmaceutical sciences, Biological sciences and/or Physical chemistry is considered a merit.
