The development of a new generation of oral killed whole cell cholera vaccines

Description of the research project
The project aims to develop a new generation of cholera vaccines to replace currently licensed killed whole cell cholera vaccines. Current vaccines require the large-scale culturing of three virulent strains of Vibrio cholerae batches of which are then killed by formalin or heat treatment. The new approach will not only considerably simplify the production of the vaccine by substituting the current three strains with only one, but will at the same time make manufacture safer since the new strain will not be toxic. The resulting new vaccines should thus be easier, safer and cheaper to manufacture, give rise to protective antibacterial as well as antitoxic immunity and ideally give effective protection after a single dose. If successful the idea can be extended to other killed whole cell vaccines under development.
The ultimate goals of the project are 1) to use gene technology to generate a single strain of Vibrio cholerae with attributes that will allow it to replace the multiple strains used in currently licensed killed whole cell cholera vaccines, 2) remove the need to add recombinant cholera toxin B subunit to the final vaccine, 3) improve immunogenicity, 4) demonstrate its immunogenicity and safety in animal models prior to the initiation of a phase I clinical trial and 5) finally to prepare material under GMP conditions for toxicity testing and make preparations for a phase I clinical trial in healthy Swedish volunteers. As such the project provides the basis of a wide-ranging and challenging Ph.D. program as the candidate student will be expected to participate in all areas except the preparation of a dry formulation.
