Genetic improvement of oil crops using the genome editing technique CRISPR/Cas9
Accompanied by a changing climate and a growing population, we are facing major challenges in supply of food, industrial raw materials and energy to meet an increasing demand for bio-based raw materials while making plant production more environmentally friendly and energy-efficient. At the Department of Plant Breeding, SLU Alnarp, we perform research, education and innovative extension work that are of great strategic importance for solving the abovementioned challenges. We use a wide range of both conventional and modern breeding technologies, ranging from field experiments and evaluation of genetic diversity to genetic, omics-, biotechnological, biochemical and physiological approaches. We are now recruiting a motivated PhD student to the Department of Plant Breeding, SLU in Alnarp.

Short project description and duties

The PhD student will work with genetic improvement of target traits (such as reduction of glucosinolate levels in seeds, increasing of seed yield, disease resistance, oil qualities or quantities, early flowering, herbicide resistance, etc. Will mainly focus on the traits with already characterized target genes), in oil crops using the latest CRISPR/Cas9 technique. The target crops will be rapeseed and field cress (Lepidium campestre) (one species will likely be focused depending on progress). Lepidium is a wild oilseed species that we at SLU have been domesticating in the past 20 years for developing it into a new oil crop, especially for Nordic countries with cold climates. It is a biennial species, very winter hardy, and has been shown to be suitable as a cover crop for reducing nutrient leaching. In Sweden, mutation lines generated by CRISPR/Cas 9 without integration of any T-DNA is considered as non-GM and field trials with such materials can be carried out without any permission as required for GM-lines. So, in this PhD project, the focus will be generating mutation lines through transient expression in protoplasts. The challenging step here is the regeneration of protoplasts into new shoots. Our lab has been establishing the protoplast regeneration protocols for both rapeseed and Lepidium in the last year with good progress.

Relevant facilities and competence in the group and department

We have all facilities in the department for performing the genome editing work including a robust pre-screeningonbsp;method of mutation lines before sending for sequencing for final confirmation of mutations. There will be at least one postdoc working on this project together with the PhD student, at least for the first two years. Meanwhile, we have also postdocs and researchers working with genome editing using CRISPR in barley and potato in the group andonbsp;the department, so we have extensive experience and strong competence in this area. The PhD student will work with the whole process from preparing theonbsp;CRISPR vectors,onbsp;tissue cultureonbsp;and transfection and shoot regeneration, molecular analyses of mutation lines, and phenotypic evaluation of mutation lines both in vitro and ex vitro.


Kvalifikationer: We seek a highly motivated and devoted candidate with master degree in plant biology or equivalent, with competence in molecular biology and biotechnology. Experience in tissue culture and plant transformation is a merit. The applicant must be fluent in both written and spoken English.
